摘要
目的研究邻苯二甲酸二(2-乙基己基)酯(DEHP)对小鼠精母(GC-2 spd)细胞JAZF1 mRNA及TR4蛋白表达的影响。方法将GC-2 spd细胞置于含终浓度为0(溶剂对照)、50、100、200μmol/L DEHP的培养液培养24 h。采用Real-time PCR方法检测细胞JAZF1 mRNA的相对表达水平,采用Western blot检测TR4蛋白表达水平。结果与对照组比较,50、100、200μmol/L DEHP染毒组GC-2 spd细胞的JAZF1 mRNA表达升高,而200μmol/L DEHP染毒组GC-2 spd细胞的TR4蛋白表达下降,差异有统计学意义(P<0.05)。结论 DEHP对生精细胞具有生殖毒性作用,可能通过JAZF1调控孤核受体TR4的表达从而影响雄性生殖功能。
        Objective To study the effects of di-( 2-ethylhexyl) phthalate( DEHP) on the expression of JAZF1 m RNA and TR4 protein in GC-2spd cells. Methods GC-2 spd cells were exposed to the DEHP medium with the final concentrations of 0,50,100,200 μmol/L,respectively and then cultured for 24 h. Real-time PCR was used to measure m RNA expression,and Western blot was used to dectect protein expression. Results Compared with the control group,the m RNA expression levels of JAZF1 in 50,100,200 μmol/L DEHP groups were increased,and the protein expression of TR4 in 200 μmol/L DEHP group was decreased,with statistically significant differences( P <0. 05). Conclusion DEHP has reproductive toxic effects on spermatogenic cells,which may regulate the expression of TR4 by the JAZF1 and thus affect the male reproductive function.
