摘要
目的观察高抵抗素血症对自发性高血压大鼠肥厚心肌中葡萄糖转运体4(GLUT4)表达的影响。方法将36只10周龄高血压大鼠(雄性),随机平均分为病毒组及对照组(每组各18只)。其中病毒组经尾静脉连续注射载有抵抗素基因的重组腺病毒,随机平均分为第1周组、第4周组、第8周组;而对照组则连续经尾静脉注射等体积生理盐水后,随机平均分为第1周组、第4周组、第8周组,分别于对应周数处死,测定各组大鼠空腹胰岛素水平,计算左室重量指数,HE染色观察心肌细胞及排列结构改变,ELISA试剂检测大鼠抵抗素水平,Western blot检测心肌组织中GLUT4的表达。结果病毒组大鼠血清抵抗素及胰岛素水平在第1周及第4周较对照组明显升高(P<0.05);左室重量指数在第4周及第8周较对照组明显升高(P<0.05);GLUT4水平在第1周、第4周及第8周中均较对照组明显降低,左室重量指数与GLUT4水平呈负相关。结论高抵抗素水平可以导致自发性高血压大鼠左室肥厚的发生,其机制可能与下调心肌中GLUT4的表达有关。
        Objective To observe the effect of high resistinemia on the expression of glucose transporter4(GLUT4) in hypertrophic myocardium of spontaneously hypertensive rats. Methods Thirty-six 10-week-old hypertensive rats(male) were randomly divided into a virus group and a control group(18 in each group). The virus group was continuously injected with the recombinant adenovirus carrying the resistin gene through the tail vein,while the control group was continuously injected with the same volume of normal saline through the tail vein. The virus group and the control group were randomly divided into the first week group, the fourth week group, and the eighth week group, respectively, and sacrificed in the corresponding weeks. The fasting insulin level of each group was measured, the left ventricular mass index(LVMI) was calculated, the cardiomyocytes and arrangement were changed by HE staining, the resistin level was detected by ELISA, and the expression of GLUT4 in myocardial tissue was detected by Western blot. Results Serum resistin and insulin levels in the virus group were significantly higher in the first week and the fourth week than in the control group(P<0.05). LVMI was significantly higher in the 4 th week and the 8 th week than in the control group(P<0.05). GLUT4 levels were significantly lower in the first week, the fourth week, and the eighth week than in the control group, and LVMI was negatively correlated with GLUT4 levels. Conclusion High level of serum resistin could cause left ventricular hypertrophy in spontaneous hypertension rats, which may be related with down-regulation of GLUT4 levels in myocardium.
