犬乳腺肿瘤组织P53基因的表达

摘要
为了提升宠物临床肿块性疾病的鉴别诊断水平,在分子生物学水平上通过对P53基因的分析对犬乳腺肿块性疾病进行应用研究。实验对6只健康标准比格犬和8例雌性犬恶性乳腺肿瘤样本利用实时荧光定量PCR(real time Q-PCR)技术定量测定P53基因的表达水平。实时荧光定量PCR结果显示,阴性对照组中,P53基因与GAPDH基因的CT值结果均大于12.5,实验组中,P53基因与GAPDH基因的CT值结果均小于10;结果分析表明,恶性乳腺肿瘤组织中P53基因表达量与正常乳腺组织中的P53基因表达量差异均显著,其中3号差异显著(☆:P<0.05),其余差异均极显著(☆☆以上:P<0.01)。结果表明:P53基因表达水平可作为未来宠物临床监测犬乳腺肿块疾病的一项重要分子水平指标。该研究结果对犬乳腺肿瘤分子生物学诊断的发展提供参考。
In order to enhance the level of differential diagnosis of tumor disease in pet clinical,make the application research of canine mammary tumor disease through the analysis of P53 gene on the level of molecular biology.We select 6 healthy standard beagle and 8 cases of female dog malignant breast tumor samples to quantitative determination of P53 gene expression level by real-time quantitative PCR(real time Q- PCR)technology.The determination result of real time Q-PCR shows that the result of CT value of P53 gene and GAPDH gene are greater than 12.5 in the group of negative,the result of CT value of P53 gene and GAPDH gene are less than10 in the experimental group.The analysis of result shows that P53 gene expression in malignant breast tumor tissue volume and P53 gene expression in normal breast tissue volume are significantly difference,the number of three is significant difference(☆:P<0.05),the remaining differences were very significant(☆☆以上:P<0.01).The result shows that P53 gene expression level can be used as an important molecular index of monitoring breast lumps disease in pet dogs clinical in the future.The result of research provide the reference on the development of molecular biology diagnosis to canine breast tumor.
