Screening and Identifying Novel Resistance Sources to Stripe Rust of Wheat

Stripe rust,caused by Puccinia striiformis Westend.f.sp.tritici Erikss,is one of the most damaging diseases in the world.To identify new sources of resistance and the resistance genes to stripe rust,120 wheat entries were assessed for stripe rust resistance at adult plant stage in Yangling,Shaanxi province artificially inoculated and in Tianshui,Gansu province naturally infected from 2010 to2015 in field test.Nine races including current prevalent stripe races(CYR32,CYR33,V26),potential prevalent races(Sull-4,Sull- 5,Sull- 7) and past prevalent stripe races(CYR23,CYR29,CYR31) were inoculated separately at seedling stage in the greenhouse.Presence or absence of known resistance genes were postulated with molecular marker linked with Yr5,Yr9,Yr10,Yr15,Fr17,Fr18,Yr26,combining with resistance spectra.The result showed 58 wheat entries had adult-plant resistance to stripe rust almost all tested years.Among these,2 entries including Xinan314-1 and Mulan were resistant to all tested races in seedlings.Molecular test implied that Xinan314-1 and Mulan did not include any tested genes,however,due to their wide-spectra resistance,it was speculated that the 2entries might have novel unknown all-stage resistance genes.Two entries including LinY867 and Bouquet had seedling susceptibility to all tested races,molecular test showed that LinY867 and Bouquet do not include any tested genes.It was speculated that they might have unknown adult-plant resistance genes.Fifty-four entries were resistant to at least one tested race,indicating they might have race-specific resistance.Among these,Bainong160 included Yr17,Zhoumai17,Shaanmai536 included Yr9,Annong1401 included Yr26.Datang991,Qian15,Yeereryan-1-3 include Yr9 + Yr17,Annong1236,Xinong418 and Jiumai2 included Yr9 + Yr26,the remaining entries did not include any tested genes,these 47 entries might have unknown genes or several QTLs.It was concluded that adult-plant resistance appears to be the most effective type of resistance protecting wheat from stripe rust currently.
