针刺对急性脑损伤与神经元可塑性影响的研究

摘要
目的:研究针刺对急性脑损伤的疗效及作用机理,探讨早期
    针刺的可行性及其对脑损伤后神经再生修复及神经元可塑性的影
    响。
     方法:利用液压致伤装置,复制大鼠中度脑损伤模型:选取
    行走试验和平衡试验对损伤后1-14d行为功能障碍进行动态观
    测,同时计算死亡率:观察脑表面血肿范围,HE染色并计算损
    伤后1d伤侧皮层及海马神经元坏死率;干湿重法测脑水肿高峰
    期含水量;采用免疫组化ABC法测脑内bFGF阳性反应:用原
    位杂交的方法显示SS mRNA及GAP-43 mRNA基因表达的变化。
    并分别选用t检验、X~2检验及秩和检验进行统计学处理。
     结果:脑损伤后大鼠出现明显的行为学障碍,针刺能促进运
    动功能的恢复;而且损伤后1h针刺组作用明显优于伤后3d治疗
    组;针刺能减少伤侧皮层及海马的继发性神经元坏死;针刺使脑
    内bFGF免疫阳性神经元阳性反应增强、GAP-43 mRNA基因表
    达上调,并能影响损伤后SS mRNA异常表达;但针刺对脑血肿
    范围、脑水肿高峰期及死亡率无显著影响。
     结论:本实验研究分别从整体到基因水平讨论了针刺对急性
    脑损伤的影响,并将行为学评价与形态学研究相结合,肯定了针
    刺对脑损伤的疗效,展示了针刺能减轻脑损伤后病理改变、改善
    受损神经元再生微环境、促进脑功能重建的作用机制。
Object: To study the effect of acupuncture therapy on acute brain injury and to
     investigate the feasibility of the acupuncture in the early stage and its?influence on the
     never regeneration, reparation and neuronal plasticity following injury.
    
    
     Methods: Using fluid-percussion injury device, copy the model of the moderate brain
     injury of the rats. Select walking and balance tests to observe the behavioral obstacle from
     first day to 14th days after the injury dynamically~ meanwhile calculate the death rate; look
     into hematoma extend on the cerebral surface, HE stain and calculate the putrescence rate
     of the injured side cortex and hippocampus on the first day after the injury; measure the
     peak volume of the brain edema : Study the immunity reaction of bFGF protein by
     using ABC way of immunohistochemistry in the brain : Show SS mRNA and GAP-43
     mRNA gene expressive variety by using in situ hybridization methods. and carry through
     statistic management by choosing the T test ,Chi-square test and Rank Sum test
     separately
    
     Result: After the brain injury, the rats have obvious active obstacle, acupuncture
     therapy can promote the resume of the movement function ; furthermore , the
     acupuncture will be better effect within lh than 3d after brain injury; Acupuncture can
     reduce secondary neuronal putrescence of the injured side cortex and the hippocampus;
     Acupuncture can increase immune reaction of bFGF in the brain, induce GAP-43 mRNA
     gene expression adjust upward, and it can affect abnormal expression of SS mRNA after
     the injury; However, acupuncture has no significant effect to the range of the brain
     hematoma, brain edema and death rate.
    
     Conclusion: This experiment discuss the acupuncture's influence to the acute brain
     injury from the general level to gene level separately, and combine the active function
     observation with morphology research, confirm the acupuncture's curative effect to the
     brain injury; Lay out that acupuncture can relieve the pathologic course after the brain
     injury, improve the regenerative environment of the injured neuronal, advance the
     functional mechanism of the reconstruction of the brain function.
