OT和GnRH在妊娠奶山羊下丘脑—垂体—卵巢轴中的共存

摘要
为了探讨OT和GnRH是否在下丘脑的同一细胞中共存,本实验采用免疫组化双标记法在妊娠期奶山羊下丘脑中对OT和GnRH的分布进行了检测。结果显示:室旁核、视上核、视前交叉上核、弓状核、下丘脑外侧区、乳头体内侧核、乳头体后核等18个核团(区)有OT-GnRH免疫反应阳性双标记细胞。妊娠晚期的OT-GnRH双标细胞较早期的有显著增加(p=0.032<0.05),中期较早期变化不十分明显(p=0.064>0.05)。其中妊娠早期双标记细胞约占总阳性细胞数的49.2%,OT细胞29.3%,GnRH细胞21.5%;妊娠中期双标记细胞约占总阳性细胞数的50.5%,OT细胞27.4%,GnRH细胞17.1%;妊娠晚期双标记细胞约占总阳性细胞数的56.6%,OT细胞24.7%,GnRH细胞18.7%。结果提示:GnRH与OT在上述核团的某些细胞存在共存现象,提供了GnRH与OT在下丘脑中相互调节的形态学依据。
     为了探讨OT和GnRH是否在垂体的同一细胞中共存,本实验采用免疫组化双标记法在妊娠期奶山羊垂体中对OT和GnRH的分布进行了检测。结果显示:在垂体里,没有发现OT-GnRH免疫双阳性细胞。结果提示:OT和GnRH在垂体里不发生直接调控。OT可能与腺垂体中的GnRH受体作用。
     为了探讨OT和GnRH是否在黄体的同一细胞中共存,在妊娠期奶山羊卵巢黄体中本实验采用免疫荧光双标记法结合激光扫描共聚焦显微镜观察对OT与GnRH的分布进行了检测。结果显示:在不同妊娠周期中,均观察到了OT-GnRH免疫反应双阳性细胞。妊娠中期的OT-GnRH双阳性细胞比早期显著增加(p=0.021<0.05),到了妊娠晚期,双阳性细胞逐渐恢复到早期水平(p=0.053>0.05)。妊娠早期双标记细胞约占总阳性细胞数的5.3%,OT细胞58.4%,GnRH细胞36.3%;妊娠中期双标记细胞约占总阳性细胞数的18.8%,OT细胞25.9%,GnRH细胞55.3%;妊娠晚期双标记细胞约占总阳性细胞数的10.6%,OT细胞71.6%,GnRH细胞17.8%。结果提示:OT-GnRH共同存在于黄体某些细胞中,提供了GnRH与OT在卵巢黄体中相互调节的形态学依据。
In the present study, double-label immunohistochemistry were adopted to explore co-localization of OT and GnRH in cells of hypothalamus of dairy goat during pregnancy. The results showed that neurons which were double positive with both antigens were localized in nuclei such as paraventricular nucleus, supraoptic nucleus, suprachiasmatic nucleus, arcuate nucleus, lateral hypothalamic area, medial mammillary nucleus and posterior mammillary nucleus. The number of these neurons was significantly changed during different period of gestation. From early to late pregnancy, the number of OT-GnRH immunoreactive positive neurons striking increases(p=0.032<0.05), On intermediate stage the change in the number of OT-GnRH immunoreactive positive neurons isnot statistically(p=0.064>0.05).These results provide morphological evidence for a directmodulation between GnRH and OT systms.

The result provide morphological evidence for a indirect modulation between GnRH and OT system, which OT maybe conjugate GnRH receptor.
     The coexistence of oxytocin and gonadotropin-releasing hormone immunoreactivityneurons were investigated in ovary goats using double lable immunohistochemistry combined with confocal laser scanning microscopy. The results showed that neurons whichwere double positive with both antigens were found in corepus luteum ovary. From early to intermediate pregnancy, the number of OT-GnRH immunoreactive positive neurons striking increases(p=0.021<0.05), On late stage the change in the number of OT-GnRH immunoreactive positive neurons is not statistically(p=0.053>0.05).These results providemorphological evidence for a direct modulation between GnRH and OT systms.
