FGF-2和Osteopontin在非小细胞肺癌中的表达及其相关性研究

摘要
目的:
     (1)探讨成纤维细胞生长因子-2(Fibroblast Growth Factor-2,FGF-2)和骨桥蛋白(Osteopontin,OPN)在非小细胞肺癌(non-small cell lung cancer,NSCLC)组织中的表达及其临床病理学意义。
     (2)观察FGF-2对人肺腺癌细胞株A549中OPN表达的影响。方法:
     (1)应用免疫组织化学SP法分析76例非小细胞肺癌组织、15例正常肺组织中FGF-2和OPN蛋白的表达水平。
     (2)采用Western blotting检测FGF-2对A549细胞中OPN蛋白表达水平的影响。
     (3)采用逆转录-聚合酶链反应(RT-PCR)检测FGF-2对A549细胞中OPN mRNA表达水平的影响。
     结果:
     (1)在76例NSCLC组织中,50例FGF-2蛋白表达阳性,阳性率为65.8%(50/76),阳性信号主要定位于癌细胞胞浆和胞膜;15例正常肺组织中2例FGF-2蛋白表达阳性,阳性率为13.3%(2/13);两者比较差异显著(P<0.01)。FGF-2阳性表达与淋巴结转移、临床分期密切相关(P<0.01),与年龄、性别、病理分型无关(P>0.05)。
     (2)在76例NSCLC组织中,46例OPN蛋白表达阳性,阳性率为60.5%(46/76),阳性信号主要定位于癌细胞胞浆和胞膜;15例正常肺组织中无OPN蛋白表达,阳性率为0%(0/15);两者比较差异显著(P<0.01)。OPN阳性表达与淋巴结转移、临床分期密切相关(P<0.01),与年龄、性别、病理分型无关(P>0.05)。
     (3)FGF-2和OPN蛋白表达在NSCLC中呈正相关(r=0.552,P<0.01)。
     (4)FGF-2可以上调A549细胞中OPN mRNA和OPN蛋白的表达。
     结论:
     (1)FGF-2蛋白和OPN蛋白在NSCLC中高表达,且与NSCLC的侵袭转移相关。
     (2)FGF-2蛋白和OPN蛋白表达在NSCLC中呈正相关,FGF-2促进NSCLC侵袭转移的机制可能部分与上调OPN的表达有关。
Objective:
     (1)To investigate the expression of FGF-2 and OPN protein in NSCLC tissues and analyze the correlation between them.
     (2)To investigate the effect of basic Fibroblastic Growth Factor-2 (FGF-2)on the expression of OPN in NSCLC cell line A549.
     Methods:
     (1)The expression of FGF-2 and OPN protein was detected by immunohistochemistry in NSCLC tissues and normal lung tissues.
     (2) The expression of OPN protein induced by FGF-2 in A549 cells was detected by Western blotting.
     (3)The expression of OPN mRNA induced by FGF-2 in A549 cells was detected by RT-PCR.
     Results:
     (1)The FGF-2 protein was overexpressed in NSCLC tissues. The positive expression of FGF-2(65.8%) protein in 76 NSCLC tissues was significantly higher than that in 15 normal lung tissues(13.3%)(P<0.01). The expression of FGF-2 was significantly related to TNM stages and lymphnode metastasis (P<0.01),but not to histological types, sex and age of NSCLC patients(P>0.05).
     (2) The OPN protein was overexpressed in NSCLC tissues. The positive expression of OPN(60.5%)protein in 76 NSCLC tissues was significantly higher than that in 15 normal lung tissues. (0%) (P<0.01). The expression of OPN was significantly related to TNM stages and lymph node metastasis(P<0.01),but not to histological types, sex and age of NSCLC patients.(P>0.05).
     (3)A positive correlation was found between the expression of FGF-2 and OPN protein in NSCLC(r=0.552,P<0.01).
     (4) The expression of OPN mRNA and protein was up-regulated in A549 cells when it was treated with FGF-2.
     Conclusions:
     (1)The overexpression of FGF-2 and OPN was related with the metastasis and invasion of NSCLC.
     (2) A positive correlation was found between the expression of FGF-2 and OPN in NSCLC.FGF-2 might promote the metastasis and invasion of NSCLC depending on upregulation of OPN expression.
